Die Enantiomerenverhältnisse und die Absolutmengen des chiralen Inhalationsanästhetikums Desfluran wurden in Blut und Urin von Patienten, die sich einer Narkose mit dem Racemat unterzogen hatten, mittels Headspace-Gaschromatographie-Massenspektrometrie bestimmt. Es wurde eine unterschiedliche Pharmakokinetik der Desfluran-Enantiomere aufgrund enantioselektiver Verteilung im Organismus festgestellt. Die Enantiomere von Compound B (2-(Fluormethoxy)-3-methoxy-1,1,1,3,3-pentafluorpropan), ein chirales Zersetzungsprodukt des Inhalationsanästhetikums Sevofluran, wurden gaschromatographisch an verschiedenen Cyclodextrinphasen getrennt. Dabei wurde der höchste chirale Trennfaktor, der bislang beschrieben worden ist, gefunden. Die thermodynamischen Daten der Enantiomerentrennung wurden bestimmt. Zusätzlich konnten die Enantiomere von Compound B durch präparative GC isoliert und die Absolutkonfigurationen durch anomale Röntgenbeugung bestimmt werden.
The enantiomeric ratio of the chiral inhalation anesthetic desflurane was determined in blood and urine of patients undergoing anesthesia with the racemic mixture. The headspace gas chromatography-mass spectrometry method was used. A different pharmacokinetic of the enantiomers due to enantioselective distribution was found. Furthermore, the enantiomer separation of Compound B (2-(fluoromethoxy)-3-methoxy-1,1,1,3,3-pentafluoropropane), a chiral degradation product of the inhalational anesthetic sevoflurane, was performed using different cyclodextrin derivatives as chiral selectors. The highest separation factor thus far described in enantioselective GC was found. The thermodynamic data of the enantiomer separation were determined. Additionally, the enantiomers of Compound B were isolated by preparative GC and their absolute configurations were determined by X-ray crystallography.
